D'Alessio followed her first-round 67 with another steady round Friday to lead Australia's Sarah Kemp (67). D'Alessio had a two-round total of 9-under 135.
South Africa's Ashleigh Simon (7) and overnight leader Marianne Skarpnord (73) of Norway were three shots behind.
Lorie Kane of Canada shot the day's best round of 66 to move into fifth place, four strokes behind, while British veteran Laura Davies had 71 to remain in a tie for sixth.
D'Alessio had seven birdies, a bogey and a double bogey in her second round, carding 33 on her outward nine and coming home in 35.
Kemp's round at the par-72 Pegasus Golf Course included seven birdies and two bogeys.
The tournament is co-sanctioned by the Australasian and European tours.
